Inhibition of carcinogen induced c-Ha-ras and c-fos proto-oncogenes expression by dietary curcumin
BACKGROUND: We investigated the chemopreventive action of dietary curcumin on 7,12-dimethylbenz(a)anthracene (DMBA)-initiated and 12,0-tetradecanoylphorbol-13-acetate (TPA)-promoted skin tumor formation in Swiss albino mice. Curcumin, a yellow coloring matter isolated from roots of Curcuma longa Linn, is a phenolic compound possessing antioxidant, free radical scavenger, and antiinflammatory properties. It has been shown by previously reported work that TPA-induced skin tumors were inhibited by topical application of curcumin, and curcumin has been shown to inhibit a variety of biological activities of TPA. Topical application of curcumin was reported to inhibit TPA-induced c-fos, c-jun and c-myc gene expression in mouse skin. This paper reports the effects of orally administered curcumin, which was consumed as a dietary component at concentrations of 0.2 % or 1 %, in ad libitum feeding. RESULTS: Animals in which tumors had been initiated with DMBA and promoted with TPA experienced significantly fewer tumors and less tumor volume if they ingested either 0.2% or 1% curcumin diets. Also, the dietary consumption of curcumin resulted in a significantly decreased expression of ras and fos proto-oncogenes in the tumorous skin, as measured by enhanced chemiluminesence Western blotting detection system (Amersham). CONCLUSIONS: Whereas earlier work demonstrated that topical application of curcumin to mouse skin inhibited TPA-induced expression of c-fos, c-jun and c-myc oncogenes, our results are the first to show that orally consumed curcumin significantly inhibited DMBA- and TPA-induced ras and fos gene expression in mouse skin
On the Trace Anomaly and the Anomaly Puzzle in N=1 Pure Yang-Mills
The trace anomaly of the energy-momentum tensor is usually quoted in the form
which is proportional to the beta function of the theory. However, there are in
general many definitions of gauge couplings depending on renormalization
schemes, and hence many beta functions. In particular, N=1 supersymmetric pure
Yang-Mills has the holomorphic gauge coupling whose beta function is one-loop
exact, and the canonical gauge coupling whose beta function is given by the
Novikov-Shifman-Vainshtein-Zakharov beta function. In this paper, we study
which beta function should appear in the trace anomaly in N=1 pure Yang-Mills.
We calculate the trace anomaly by employing the N=4 regularization of N=1 pure
Yang-Mills. It is shown that the trace anomaly is given by one-loop exact form
if the composite operator appearing in the trace anomaly is renormalized in a
preferred way. This result gives the simplest resolution to the anomaly puzzle
in N=1 pure Yang-Mills. The most important point is to examine in which scheme
the quantum action principle is valid, which is crucial in the derivation of

Room temperature plasmon laser by total internal reflection
Plasmon lasers create and sustain intense and coherent optical fields below
light's diffraction limit with the unique ability to drastically enhance
light-matter interactions bringing fundamentally new capabilities to
bio-sensing, data storage, photolithography and optical communications.
However, these important applications require room temperature operation, which
remains a major hurdle. Here, we report a room temperature semiconductor
plasmon laser with both strong cavity feedback and optical confinement to
1/20th of the wavelength. The strong feedback arises from total internal
reflection of surface plasmons, while the confinement enhances the spontaneous

Coherent optical wavelength conversion via cavity-optomechanics
We theoretically propose and experimentally demonstrate coherent wavelength
conversion of optical photons using photon-phonon translation in a
cavity-optomechanical system. For an engineered silicon optomechanical crystal
nanocavity supporting a 4 GHz localized phonon mode, optical signals in a 1.5
MHz bandwidth are coherently converted over a 11.2 THz frequency span between
one cavity mode at wavelength 1460 nm and a second cavity mode at 1545 nm with
a 93% internal (2% external) peak efficiency. The thermal and quantum limiting
noise involved in the conversion process is also analyzed, and in terms of an
equivalent photon number signal level are found to correspond to an internal

Mycolactone-dependent depletion of endothelial cell thrombomodulin is strongly associated with fibrin deposition in Buruli ulcer lesions
A well-known histopathological feature of diseased skin in Buruli ulcer (BU) is coagulative necrosis caused by the Mycobacterium ulcerans macrolide exotoxin mycolactone. Since the underlying mechanism is not known, we have investigated the effect of mycolactone on endothelial cells, focussing on the expression of surface anticoagulant molecules involved in the protein C anticoagulant pathway. Congenital deficiencies in this natural anticoagulant pathway are known to induce thrombotic complications such as purpura fulimans and spontaneous necrosis. Mycolactone profoundly decreased thrombomodulin (TM) expression on the surface of human dermal microvascular endothelial cells (HDMVEC) at doses as low as 2ng/ml and as early as 8hrs after exposure. TM activates protein C by altering thrombin's substrate specificity, and exposure of HDMVEC to mycolactone for 24 hours resulted in an almost complete loss of the cells' ability to produce activated protein C. Loss of TM was shown to be due to a previously described mechanism involving mycolactone-dependent blockade of Sec61 translocation that results in proteasome-dependent degradation of newly synthesised ER-transiting proteins. Indeed, depletion from cells determined by live-cell imaging of cells stably expressing a recombinant TM-GFP fusion protein occurred at the known turnover rate. In order to determine the relevance of these findings to BU disease, immunohistochemistry of punch biopsies from 40 BU lesions (31 ulcers, nine plaques) was performed. TM abundance was profoundly reduced in the subcutis of 78% of biopsies. Furthermore, it was confirmed that fibrin deposition is a common feature of BU lesions, particularly in the necrotic areas. These findings indicate that there is decreased ability to control thrombin generation in BU skin. Mycolactone's effects on normal endothelial cell function, including its ability to activate the protein C anticoagulant pathway are strongly associated with this. Fibrin-driven tisischemia could contribute to the development of the tissue necrosis seen in BU lesions
The role of mutation rate variation and genetic diversity in the architecture of human disease
Background
We have investigated the role that the mutation rate and the structure of genetic variation at a locus play in determining whether a gene is involved in disease. We predict that the mutation rate and its genetic diversity should be higher in genes associated with disease, unless all genes that could cause disease have already been identified.
Results
Consistent with our predictions we find that genes associated with Mendelian and complex disease are substantially longer than non-disease genes. However, we find that both Mendelian and complex disease genes are found in regions of the genome with relatively low mutation rates, as inferred from intron divergence between humans and chimpanzees, and they are predicted to have similar rates of non-synonymous mutation as other genes. Finally, we find that disease genes are in regions of significantly elevated genetic diversity, even when variation in the rate of mutation is controlled for. The effect is small nevertheless.
Conclusions
Our results suggest that gene length contributes to whether a gene is associated with disease. However, the mutation rate and the genetic architecture of the locus appear to play only a minor role in determining whether a gene is associated with disease
Radiation-pressure cooling and optomechanical instability of a micro-mirror
Recent experimental progress in table-top experiments or gravitational-wave
interferometers has enlightened the unique displacement sensitivity offered by
optical interferometry. As the mirrors move in response to radiation pressure,
higher power operation, though crucial for further sensitivity enhancement,
will however increase quantum effects of radiation pressure, or even jeopardize
the stable operation of the detuned cavities proposed for next-generation
interferometers. The appearance of such optomechanical instabilities is the
result of the nonlinear interplay between the motion of the mirrors and the
optical field dynamics. In a detuned cavity indeed, the displacements of the
mirror are coupled to intensity fluctuations, which modifies the effective
dynamics of the mirror. Such "optical spring" effects have already been
demonstrated on the mechanical damping of an electromagnetic waveguide with a
moving wall, on the resonance frequency of a specially designed flexure
oscillator, and through the optomechanical instability of a silica
micro-toroidal resonator. We present here an experiment where a
micro-mechanical resonator is used as a mirror in a very high-finesse optical
cavity and its displacements monitored with an unprecedented sensitivity. By
detuning the cavity, we have observed a drastic cooling of the micro-resonator
by intracavity radiation pressure, down to an effective temperature of 10 K. We
have also obtained an efficient heating for an opposite detuning, up to the
observation of a radiation-pressure induced instability of the resonator.
Further experimental progress and cryogenic operation may lead to the
experimental observation of the quantum ground state of a mechanical resonator,
either by passive or active cooling techniques
Clinical characteristics and prognosis of acute bacterial meningitis in elderly patients over 65: a hospital-based study
<p>Abstract</p> <p>Background</p> <p>To examine the clinical characteristics of bacterial meningitis in elderly patients.</p> <p>Methods</p> <p>261 patients with adult bacterial meningitis (ABM), collected during a study period of 11 years (2000-2010), were included for study. Among them, 87 patients aged ≥ 65 years and were classified as the elderly group. The clinical and laboratory characteristics and prognostic factors were analyzed, and a clinical comparison with those of non-elderly ABM patients was also made.</p> <p>Results</p> <p>The 87 elderly ABM patients were composed of 53 males and 34 females, aged 65-87 years old (median = 71 years). Diabetes mellitus (DM) was the most common underlying condition (34%), followed by end stage renal disease (7%), alcoholism (4%) and malignancies (4%). Fever was the most common clinical manifestation (86%), followed by altered consciousness (62%), leukocytosis (53%), hydrocephalus (38%), seizure (30%), bacteremia (21%) and shock (11%). Thirty-nine of these 87 elderly ABM patients had spontaneous infection, while the other 48 had post-neurosurgical infection. Forty-four patients contracted ABM in a community-acquired state, while the other 43, a nosocomial state. The therapeutic results of the 87 elderly ABM patients were that 34 patients expired and 53 patients survived. The comparative results of the clinical and laboratory characteristics between the elderly and non-elderly ABM patients showed that only peripheral blood leukocytosis was significant. Presence of shock and seizure were significant prognostic factors of elderly ABM patients.</p> <p>Conclusions</p> <p>Elderly ABM patients accounted for 34.8% of the overall ABM cases, and this relatively high incidence rate may signify the future burden of ABM in the elderly population in Taiwan. The relative frequency of implicated pathogens of elderly ABM is similar to that of non-elderly ABM. Compared with non-elderly patients, the elderly ABM patients have a significantly lower incidence of peripheral blood leukocytosis. The mortality rate of elderly ABM remains high, and the presence of shock and seizures are important prognostic factors.</p
